NAD+ is essential for energy production within cells, supports mitochondrial function, and plays a role in the creation of neurotransmitters—all of which are involved in mood and stress regulation. Certain studies propose that low NAD+ levels may be linked to anxiety symptoms, especially in cases involving chronic fatigue or long-term stress.
A study by Alegre and colleagues in 2010 explored oral NADH supplements in patients with chronic fatigue syndrome. The double-blind, placebo-controlled trial showed a statistically significant reduction in anxiety scores for the NADH group (p < 0.05). While the participants had chronic fatigue, the observed decrease in anxiety proposes a potential benefit for those experiencing similar symptoms.
A 2024 systematic review published in the American Journal of Physiology examined multiple randomized clinical trials. It found that NADH and NAD+ precursor supplements were generally well tolerated and associated with lower anxiety symptoms and better quality of life outcomes.
Important context: Most of the current research focuses on people with chronic fatigue or studies where anxiety was a secondary outcome.
